北卡罗莱纳大学威尔明顿分校的前身是开办于1947年9月4日的威尔明顿学院。1963年7月1日威尔明顿学院成为一所可以提供四年制本科学位课程的高级学院。1969年7月1日威尔明顿学院更名为北卡罗莱纳大学威尔明顿分校。
大学下设文理学院、卡梅伦商学院、沃森教育学院和卫生护理学院，提供四年制的本科学位课程，包括文理学士、音乐学士以及社会工作学士。大学研究生院还可获颁文理硕士、教学文科硕士、工商行政管理硕士、教育学硕士、创意写作美术硕士、学校管理硕士、会计学理科硕士和护理学理科硕士。大学的强项是自然科学，特别是生物科学、化学和海洋科学。创意写作美术硕士课程已成为东部海岸地区最受尊敬和最具竞争力的课程之一。北卡罗莱纳大学威尔明顿已经第11年被美国新闻与世界报道列入能提供硕士课程的南部公立大学前10名。2009年大学的排名为第六位。在南部所有的121所公立与私立教育机构中，北卡罗莱纳大学威尔明顿排名14。
